Testicular Toxicity of Arsenic on Spermatogenesis in Fish

چکیده
Arsenic (As), a well-known water pollutant, has been associated with various human health problems, however, studies pertaining to its reproductive toxicity on fish are very limited. In this study, we investigated the direct influence of As on fish spermatogenesis using Japanese eel (Anguilla japonica) in vitro testicular organ culture system. Eel testicular fragments were cultured in vitro with increasing concentrations of As with or without human chorionic gonadotropin (hCG). Thereafter, steroid synthesis, steroidogenic enzyme activity, testicular morphology and proliferation of germ cells were analyzed. As inhibits hCG-induced germ cell proliferation in a dose-dependent manner. Highest dose of As (100 μM) with hCG strongly induced germ cell apoptosis. Moreover, a dose of As (0.1 μM) lower than the WHO drinking water quality guideline most effectively suppressed 11-ketotestosterone (11KT) synthesis. At the same As concentration, 3.-hydroxysteroid dehydrogenase (3β-HSD) expression and activity were inhibited. A high oxidative DNA damage was observed in testicular fragments exposed to the same treatment (100 μM As + hCG) which caused apoptosis. These results suggest that low dose of As inhibits spermatogenesis via suppression of steroidogenic enzyme activity and expression while high dose of this compound induces oxidative stress-mediated germ cell apoptosis.

Arsenic induced toxicity on testicular tissue of mice.
Effect of arsenic was studied on the testicular tissue of Swiss albino mice. Sodium-meta-arsenite (NaAsO2) was administered to adult mice (25 +/- 30 g) at a dose level of 30 mg/L and 40 mg/L through drinking water for 30, 45 and 60 days. After the treatment, the testicular organ was removed, weighed and processed for histopathological observation. No change in the body weight was recorded in tr...

Objective: The main objective of the current study was to observe the ameliorative effect of Withania somnifera on arsenic-induced testicular toxicity by exploring the crucial parameters such as sperm counts, sperm motility, hormonal assay and lipid peroxidation including histopathology. Materials and Methods: In the present study, arsenic in the form of sodium arsenite was administered orally ...

Arsenic has a suppressive influence on spermatogenesis and induces impairment in male reproductive system due to oxidative stress. The present study was aimed to test the arsenic induced toxicity and protection by Chlorophytum borivilianum. The effect of sodium arsenite (4 mg/(kg body weight (bw) x day)) via double distilled water without or with C. borivilianum (800 mg/(kg bw x day)) was evalu...

Background Background: Mammalian spermatogenesis is regulated through paracrine and endocrine activity, specific cell signaling, and local control mechanisms. These highly specific signaling interactions are effectively absent upon placing testicular cells into two-dimensional primary culture. The specific changes that occur between key cell types and involved spermatogenesis signaling pathways...
متن کاملEffects of chronic exposure to sodium arsenite on hypothalamo-pituitary-testicular activities in adult rats: possible an estrogenic mode of action
BACKGROUND Inorganic arsenic is a major water pollutant and a known human carcinogen that has a suppressive influence on spermatogenesis and androgenesis in male reproductive system. However, the actual molecular events resulting in male reproductive dysfunctions from exposure to arsenic remain unclear. In this context, we evaluated the mode of action of chronic oral exposure of sodium arsenite...
